However, challenges remain in generating standardized human brain organoids that can recapitulate key physiological features of a human brain. Here, we present tubular organoid-on-a-chip devices to generate better organoids and model neuroinflammation. By employing 3D printed hollow mesh scaffolds, our device can be easily incorporated into multiwell-plates for reliable, scalable, and reproducible generation of tubular organoids. By introducing rocking flows through the tubular device channel, our device can perfuse nutrients and oxygen to minimize organoid necrosis and hypoxia, and incorporate immune cells into organoids to model neuro-immune interactions. Compared with conventional protocols, our method increased neural progenitor proliferation and reduced heterogeneity of human brain organoids. As a proof-of-concept application, we applied this method to model the microglia-mediated neuroinflammation after exposure to an opioid receptor agonist. We found isogenic microglia were activated after exposure to an opioid receptor agonist (DAMGO) and transformed back to the homeostatic status with further treatment by a cannabinoid receptor 2 (CB2) agonist (LY2828360). Importantly, the activated microglia in tubular organoids had stronger cytokine responses compared to those in 2D microglial cultures. Our tubular organoid device is simple, versatile, inexpensive, easy-to-use, and compatible with multiwell-plates, so it can be widely used in common research and clinical laboratory settings. This technology can be broadly used for basic and translational applications in inflammatory diseases including substance use disorders, neural diseases, autoimmune disorders, and infectious diseases.Heroism is one of the most original concepts of Antoni Kępiński contained in his works. Due to low availability of sources of information it faces some interpretative problems. The aim of the article is to indicate heroism and concepts associated with it (heroic attitude, heroic tendency or heroic proportion) as the highest therapeutic possibility. The author thoroughly analyzed the literature on the subject both by Kępiński himself and other authors (e.g., Kokoszka) and also obtained some information from Kępiński's colleagues. He made the analysis of these contents, using the interpretative framework based on subject's superior potentiality - man's self acting and experiencing his action. This concept comes from Karol Wojtyła's realistic phenomenology, which brings a certain interpretative novelty in the form of phenomenologically framed will. As a result one can establish the essential connection between heroism and superior potentiality that is determined by the essence of freedom. The final discussion confirms the title thesis, which widens and completes the knowledge about studied therapeutic factor, which gains the new potential for therapeutic practice and its research. This work is a review work, which can put a foundation for further development of the studied subject based on the applied interpretive framework.The aim of this paper is to define the role of emotions in decision-making mechanisms in the concept of information metabolism -an original research project of the Polish psychiatrist Antoni Kępiński. Emotions constitute a subjective (first-person) expression of the subject's relation to his or her environment, resulting from the basic principles defined as biological laws; decisions made on the basis of emotional process and current body states are the foundation for information processing. This postulate anticipates further neurobiological research and has been referred to in Antonio Damasio's work. Determination of a biological role of emotions enables identification of their place in a psychotherapeutic process described by information metabolism. Emotions through their relational dimension constitute the genesis of the meaning of an individual life. Thanks to the definition of the emotional process as a source of connections between an individual and their world, whose aim is the choice of a possibly most optimal procedure, emotions may be treated as a basis for the situation of understanding. The notion of understanding used in this context in relation to the views of Kępiński relates in a substantial part to the subjective individual experiences and is theoretically juxtaposed with the notion of explaining. The proposal of interpretation of the model of Kępiński's psychiatry, based on understanding resulting from the emotional process, displays many common elements with various ways of approaching the psychiatric theories, e.g. biological psychiatry or post-psychiatry.Studies carried out so far have shown that negative stereotypes concerning people with gambling disorders and, in particular, the belief that these people bear personal responsibility for their illness, can significantly contribute to the stigmatization of these people. Shame and fear of stigmatization significantly hinder the decision to start treatment. This study investigates the beliefs of respondents about the impact of stigmatization of people with gambling disorders on the social perception of treatment of gambling disorders and the beliefs of respondents about the impact of stigmatization on undertaking treatment or seeking help. Furthermore, the study investigates whether treatment can help reduce stigma and whether professionals, in some way, take this problem into account in their practice.
In the first half of 2015, 90 semi structured individual interviews were conducted with people with gambling disorders, social workers, therapists employed in addiction treatment facilities, General Practitioners and psychiatrists.
